Fainting is due to decreased blood flow to the brain (usually), and being knocked out is trauma to the brain.
We use drugs that interrupt the normal brain activity that has to do with consciousness, as well as drugs to relax muscles, prevent/treat pain and anxiety, and other things. They work on a cellular level, and most of our drugs increase blood flow to the brain, the opposite of fainting.
